Preliminary Studies on the Establishment of Seedling Production System by Somatic Embryyogenesis in Oenanthe stolonifera DC

Abstract

|
In an effort to develop efficient seedling propagation system using somatic embryos for Oenanthe stolonifera hydroponic culture, studies have been conducted to involve the germination factors of somatic embryos. The highest rate of germination from both primary and secondary embryos was obtained in MS media supplemented either with BA 1.0 §·¡¤L^(-1) or kinetin 1.0 §·¡¤L^(-1) alone, or with NAA 0.01 §·¡¤L^(-1) and BA 0.5 §·¡¤L^(-1) combined. Shoots and roots formed best from somatic embryos in suspension cultures containing MS medium with BA 0.1 §·¡¤L^(-1) when subjected to a gentle agitation (25 rpm). Somatic embryos acclimatized either by drying at room temperature for 24 hours or by storage at 4¡É for 5 days, which failed to germinate under unsterile conditions, greatly increased the rate of germination compared to unacclimatized embryos when germinated under sterile conditions.
